Our research interests include the signal transduction pathways activated by growth factors, cytokines and hyperglycemic condition. We are interested in mechanisms that support the development of diabetic kidney disease and renal cell carcinomas. Nearly 50% of the patients with diabetes develop nephropathy. Importantly, diabetic patients with kidney disease have 87% higher risk of cardiovascular mortality when compared with those without kidney disease. The early pathologic changes in diabetic nephropathy involve renal, especially glomerular and tubular hypertrophy and expression of matrix proteins such as collagen, fibronectin and laminin. My laboratory has been studying the signal transduction mechanisms that lead to the progression of diabetic nephropathy. To test our concepts, we use renal cells (glomerular mesangial cells and proximal tubular epithelial cells) in culture and mouse and rat models of type 1 and type 2 diabetes. TGF (transforming growth factor-) mediates many pathologic effects of hyperglycemia. Therefore, we study the signaling mechanisms of this cytokine in mesangial and proximal tubular epithelial cells. We discovered that the tumor suppressor protein PTEN (phosphatase and tensin homolog deleted on chromosome 10) is downregulated in the kidneys of diabetic rodents. We established that the suppressed PTEN contributes to the pathologies of diabetic nephropathy. To identify the mechanisms that may regulate the suppression of PTEN under hyperglycemic condition, we studied several microRNAs. In last few years, we showed that miR-21, miR-26a and miR-214 control the expression of PTEN in renal cells in response to high glucose/TGF and in the kidneys of diabetic mice. These studies opened the door to the novel application of anti-microRNA therapy for diabetic nephropathy. We previously showed that mTOR plays an important role in the pathologies of diabetic nephropathy. For the pathologic effects in the disease, mTOR is activated in sustained manner. More recently, we have focused on a novel protein, called deptor, which is a component of mTOR complexes 1 and 2. In fact deptor is an endogenous inhibitor of mTOR activity. For the first time, we showed that the renal expression of deptor was significantly reduced in humans with diabetes and in diabetic rodents and that this reduction contributed to the increased mTOR activity. In cultured mesangial and proximal tubular epithelial cells incubated with high glucose or TGF, the expression of deptor was significantly reduced, which resulted in sustained activation of both mTORC1 and mTORC2. Thus, it is important to study the mechanism of deptor downregulation by hyperglycemia. We are currently investigating the epigenetic, post-transcriptional (microRNAs) and post-translational mechanisms of deptor downregulation in diabetic nephropathy.
